Abstract: Classic anatomical methods have failed to determine the precise location, origin and nature of nerve fibres in the inferior hypogastric plexus (IHP). The purpose of this study was to identify the location and nature (adrenergic and/or cholinergic) of IHP nerve fibres and to provide a three-dimensional (3D) representation of pelvic nerves and their relationship to other anatomical structures. Serial transverse sections of the pelvic portion of two human male fetuses (16 and 17 weeks' gestation) were studied histologically and immunohistochemically, digitized and reconstructed three-dimensionally. 3D reconstruction allowed a 'computer-assisted dissection', identifying the precise location and distribution of the pelvic nerve elements. Proximal (supra-levator) and distal (infra-levator) communications between the pudendal nerve and IHP were observed. By determining the nature of the nerve fibres using immunostaining, we were able to demonstrate that the hypogastric nerves and pelvic splanchnic nerves, which are classically considered purely sympathetic and parasympathetic, respectively, contain both adrenergic and cholinergic nerve fibres. The pelvic autonomic nervous system is more complex than previously thought, as adrenergic and cholinergic fibres were found to co-exist in both 'sympathetic' and 'parasympathetic' nerves. This study is the first step to a 3D cartography of neurotransmitter distribution which could help in the selection of molecules to be used in the treatment of incontinence, erectile dysfunction and ejaculatory disorders.

Abstract: BACKGROUND: Detailed knowledge of nerve distribution in the neurovascular bundle (NVB) is essential to preserve sexual function after prostatic surgery. OBJECTIVE: To identify the location as well as the type (adrenergic, cholinergic, and sensory) of nerve fibres within the NVB and to provide a three-dimensional (3D) representation of their structural relationship in the human male foetus. DESIGN, SETTING, AND PARTICIPANTS: Serial transverse sections were performed every 150-200 microm in the pelvic portion of six human male foetuses (15-20 wk of gestation). Sections were treated with histologic and immunohistochemical methods (hematin-eosin-safran, Luxol Fast Blue, immunolabelling of protein S100, vesicular acetylcholine transporter, tyrosine hydroxylase, calcitonin gene-related peptide, and substance P). The 3D pelvic reconstruction was obtained from digitised serial sections using WinSurf software. MEASUREMENTS: NVB nerve location and type were evaluated qualitatively. RESULTS AND LIMITATIONS: The 3D reconstruction allowed precise identification of pelvic organ innervation. Nerve fibres derived from the inferior hypogastric plexus followed two courses: posterior and lateral, providing cholinergic, adrenergic, and sensory innervation to seminal vesicles, vas deferens, prostate, and urethral sphincter. Cavernous nerve fibres did not strictly follow the NVB course; they were distributed at several levels, in a fanlike formation. The main limitations of this study were the limited number of specimens available due to legal restriction and the time-consuming nature of the manually performed stages in the method. CONCLUSIONS: The distribution of nerve fibres within the posterolateral prostatic NVB and the existence of mixed innervation in the posterior and lateral fibre courses at the level of the prostate and seminal vesicles give us an insight into how to minimise effects on sexual function during prostatic surgery. The 3D computer-assisted anatomic dissection represents an original method of applying anatomic knowledge to surgical technique to improve nerve preservation and decrease postoperative sexual complications.

Abstract: Small leucine-rich proteoglycans are components of the extracellular matrix of mineralized tissues. DS/CSPGs (i.e. decorin and biglycan) and KSPGs (i.e. lumican, osteoadherin and fibromodulin, or Fmod) have been found in dentin, cementum and bone. However, the role these PGs might have in promoting or inhibiting mineralization is still unclear.

Western blotting revealed that the Fmod MW was higher in bone (52kDa) compared with with dental tissues (52 and 40kDa). Strong immunostaining of Fmod was restricted to i) the distal border of secretory odontolblasts, ii) the distal junctional complex of secretory ameloblasts, iii) the stratum intermedium and there was faint staining in the predentin.

Fmod deficient (KO) tissues have collagen fibrils with a larger diameter than controls (WT). Dentin was also hypomineralized in the Fmod KO. In molars, the pulp chambers were wider in the KO compared to WT, and the observed differences in dentin formation were confirmed by X-ray microcomputed tomography. Structural defects were observed within the forming enamel in the incisor of the KO mice. Compensatory mechanisms appeared to partially hide the defects of dental tissues, most likely by DSP, DMP-1 and BSP which were over-expressed, but not by OPN which showed no differences between the WT and KO tissues.

Using the scanning electron microscopy, backscattering analysis of sections of mandibles of day 21 mice revealed larger surfaces occupied by the bone medulla in the KO. X-ray tomography revealed severe hypomineralization of bone, mostly in the distal part of the mandible of KO mice, including the condyle, coronoïd, angula and the vertical branch, The alveolar bone located in the central part was also defective, whereas changes were more subtle in the anterior (rostral) part. The bone deficiency in the KO was even more severe in 10 weeks- old mice.

Altogether, these data suggest that Fmod is a crucial factor in regulating mineralization. We delineate the differences between the three different parts of the mandibular bone, as well as differences between bone and tooth tissues.

Abstract: Post-menopausal osteoporosis is a common disorder that results from increased osteoclastic activity caused by estrogen deficiency.
Whether post-menopausal bone remodeling can alter the response to particulate debris is not known. The purpose of this study was to
evaluate the bone response to polyethylene particles in an estrogen deficient murine model. PE particles were implanted onto the
calvaria of seven wild-type mice and seven ovariectomized (OVX) mice. Calvaria from unimplanted wild-type and OVX mice served as
controls. Calvaria were harvested after 14 days. Skulls were analyzed with a high-resolution micro-CT and by histomorphometry after
staining with Stevenel blue and picrofuschine, and for tartrate-specific alkaline phosphatase. As assessed on micro-CT evaluation,
particles induced significant decrease of bone thickness in wild-type mice (p=0.04), while bone thickness was stable in OVX mice with
particles (p=0.40). The osteoclast number per mm total bone perimeter was in wild-type animals with particles 2.84+/-1.6 and 1.74+/-1.3
in OVX animals with particles (p=0.004). Mean bone loss in wild-type mice was -12+/-10% while mean bone loss was -4.7+/-0.9% in
OVX animals with particles (p=0.004).Particles induced a diminished osteolytic response in OVX mice, suggesting that estrogen
deficiency may have a protective role against particle-induced bone resorption. These important new findings may help to stimulate
clinical studies which may define criteria to better identify patients at risk to develop particle-induced osteolysis.

Abstract: Our objective was to evaluate particle-induced osteolysis both histologically and with the use of a microscanner in a murine model.

methylmehacrylate and sections were obtained. Sections were stained with Stevenel Blue and picrofuschine and a serial section stained
for tartrate-specific acid phosphatase-positive osteoclasts. Mean bone thickness (ratio bone thickness/tissue thickness), sagittal suture
area and osteoclasts number were measured. As assessed on micro-CT, particles induced a significant decrease in bone volume in PEtreated
mice, in comparison with sham animals (p<0.05). Fibrous and granulomatous scar tissue overlying the calvariae was observed
on histological sections. Quantitatively, the mean bone thickness was 60+/-9.5% (range, 42-80%) in PE-treated mice, in comparison
with 48+/-10% (range, 23-73%) (p<0.0001) in sham mice. The sagittal suture area and the osteoclasts number both considerably
increased after particles implantation. MicroCT data significantly correlated with histomorphometry. By implanting PE-particles onto mice
calvaria, we experimentally modeled a bone reaction very similar to the one observed in peri-prosthetic osteolysis. Both microCT and
histological tools allowed us to accurately evaluate particle-induced bone resorption.

Abstract: Postmenopausal osteoporosis is a common disorder that results from increased osteoclastic activity caused by estrogen deficiency. Whether postmenopausal bone remodeling can alter the response to particulate debris is unknown. The purpose of this study was to evaluate the bone response to polyethylene particles in an ovariectomized murine model. Polyethylene particles were implanted onto the calvaria of seven control mice and seven ovariectomized (OVX) mice, as compared with calvaria from sham-operated and OVX mice. Calvaria were harvested after 14 days. Skulls were analyzed with a high-resolution micro-CT and by histomorphometry after staining with Stevenel blue and picrofuschine, and for tartrate-specific alkaline phosphatase. As assessed by micro-CT, particle implantation induced a significant decrease in bone thickness in control mice, while bone thickness remained stable in OVX mice. In particle-implanted animals, the osteoclast number was 2.84 +/- 0.3 in control mice and 1.74 +/- 0.22 in OVX mice. Mean bone loss was -12% +/- 1.9% in control mice and -4.7% +/- 1.7% in OVX animals. The reduction of osteolytic response suggests that ovariectomy may have a protective role against particle-induced bone resorption.

Abstract: À la suite d’une série de découvertes faites à partir de 2003 dans le secteur du mastaba d’Akhethetep dans la nécropole de Saqqara en Égypte par la mission du musée du Louvre dirigée par Ch. Ziegler, une des momies les mieux conservées a été scannée au Caire par des collègues égyptiens pour les besoins d’un film destiné à la télévision française (France 3).
La reprise des données par l’équipe Useful Progress traitant d’imagerie médicale a permis d’assembler les différents scanners, de composer une vue complète de la momie, de déshabiller la dépouille de ses linceuls, d’étudier les procédés de momification et de faire l’étude anthropologique du corps de cette jeune femme décédée au IV’ siècle av. J.-C. (travail en cours par Sylvain Ordureau).
